Since, under certain conditions of culture, viable cells have only one nucleus ( 3 8 ) , there must be only one chromosome per genome, which agrees with the genetic evidence of one linkage group per genome. It is conceivable that the unreplicated chromosome may contain more than one copy of the genome. Indeed, an observation compatible with such a notion has been made by Fulton ( 2 0 ) , who found that a second round of marker transfer begins, under conditions of prolonged conjugation, with the conservation of genetic linkage across the Hfr site.
For a single marker, only 2 to 3% carry all four linked markers. In Hemo philus influenzae, Berns and Thomas ( 6 ) observed that high-molecularweight DNA preparations showed linkage between markers that were previously observed not to be linked.
